Pigmentation was determined by extraction with organic solvents and measured as optical density at 475 mμ for M. radiodurans and S. lutea, and 410 mμ for Ps E aetuginosa. Pigments of M. radiodurans and S. lutea were inactivated at a linear rate with increased doses. The non -carotenoid pigment of Ps. aeruginosa was not significantly affected at a maximum dose of 2, 040 krads. Quantitation of neutral lipids and neutral lipid hydrolysates was accomplished with thin layer chromatography. The lipid classes in -. volved were separated on thin layer plates, charred with concentrated H2SO4 and heating, and densitometric determinations made. 6s. W. No differences were observed in the lipid classes of M. radiodurans and S. lutea following doses up to 2, 040 krads. A dose of 2, 040 krads resulted in a large increase in free fatty acids of Ps. aeruginosa. A model for repair of radiation-induced DNA double-strand breaks in the extreme radiophile Deinococcus radiodurans.
The bacterium Deinococcus (formerly Micrococcus) radiodurans and other members of the eubacterial family Deinococaceae are extremely resistant to ionizing radiation and many other agents that damage DNA. Stationary phase D. radiodurans exposed to 1.0-1.5 Mrad gamma-irradiation sustains > 120 DNA double-strand breaks (dsbs) per chromosome; these dsbs are mended over a period of hours with 100% s...

The paracrystalline outer membraneous layer (HPI layer) of Micrococcus radiodurans has been investigated by negative- and positive-staining electron microscopy and subsequent digital image processing. The subunit structure of the major HPI layer protein complex and the lipid-protein distribution in the plane of the membrane have been determined. The HPI layer was found to be highly asymmetric i...
متن کاملRestorative Effect of Vitamin E Supplementation on Hepatic Lipid Peroxidation and Lipid Profile Changes Induced by Sublethal γ-Radiation in BALB/c Mice
In order to find a prophylactic supplementation for individuals who are at risk of exposure to ionizing radiation, we attempted to evaluate the effect of Vitamin E (Vit-E), a biological free radical scavenger, on restoration of hepatic lipid peroxidation (LPO) and lipid profile (LP) changes induced by sublethal g- radiation in BALB/c mice. The concentrations of cholesterol and phospholipid were...
متن کاملRepair of ultraviolet radiation damage in sensitive mutants of Micrococcus radiodurans.
Various aspects of the repair of ultraviolet (UV) radiation-induced damage were compared in wild-type Micrococcus radiodurans and two UV-sensitive mutants. Unlike the wild type, the mutants are more sensitive to radiation at 265 nm than at 280 nm. The delay in deoxyribonucleic acid (DNA) synthesis following exposure to UV is about seven times as long in the mutants as in the wild type. All thre...
متن کاملEffect of physiological age on radiation resistance of some bacteria that are highly radiation resistant.
Physiological age-dependent variation in radiation resistance was studied for three bacteria that are highly radiation resistant: Micrococcus radiodurans, Micrococcus sp. isolate C-3, and Moraxella sp. isolate 4. Stationary-phase cultures of M. radiodurans and isolate C-3 were much more resistant to gamma radiation than were log-phase cultures. This pattern of relative resistance was reversed f...
